α-synuclein (αSyn) seeding is key for spreading Parkinson’s disease (PD) pathology throughout the brain. Recently, we developed the seed amplification assay (SAA) to reproduce αSyn seeding in vitro with high efficiency. SAA offers a great opportunity to detect αSyn seeds in human samples and to understand the disease biology. The main goal of this project is to understand αSyn seeding in the context of Parkinson’s disease, by identification of cellular factors modulating seeding, evaluating the nature of αSyn seeds detected by SAA in diverse tissues and fluid samples, and determining the structural and biological relevance of the SAA products.
