Deciphering the Aggregatome: Unraveling Alpha-Synuclein Seeding Drivers in Parkinson’s Disease across Human Biomaterials

Parkinson’s disease progression varies significantly among patients, with alpha-synuclein (aSyn) aggregation as a key pathological hallmark. However, factors influencing aSyn seeding remain poorly understood. We will characterize biochemical and biophysical determinants of aSyn seeding across brain, CSF, blood, skin and stool. Our multidisciplinary team brings together expertise in seed amplification assay development, mass spectrometry-based proteomics, advanced biochemical methods, neuropathological characterization and access to exclusive patient cohorts with longitudinally matched biosamples. We will: 1) define differentially seeding-competent aSyn species and their molecular signatures; 2) validate their functional relevance in cellular models; 3) establish their association with disease progression and clinical phenotypes.
